Liam’s research project aims to leverage environmental DNA (eDNA) alongside traditional freshwater macroinvertebrate community and water quality assessment methods, combining each approach into a holistic ecological health assessment of the River Ely. Applied eDNA assessments will be used as a non-invasive and high sensitivity tool investigating the presence and distribution of the cryptic and highly invasive amphipods Dikerogammarus haemobaphes (Demon shrimp) and Dikerogammarus villosus (Killer shrimp), utilising DNA barcoding techniques to amplify trace species-specific genetic material via polymerase chain reaction (PCR) within sediment samples taken from Cardiff Bay and several sites along the River Ely. The results will supplement preserved macroinvertebrate kick samples identified to the lowest taxonomic rank possible, in order to determine the range of these INNS within the Ely catchment whilsts assessing the river’s current macroinvertebrate community composition. The results of this research aim to demonstrate the efficacy of eDNA as a rapid, scalable and cost effective monitoring tool to identify invasive species that may be challenging to detect through conventional sampling methods, highlighting its potential as a supplemental tool alongside traditional assessment methods, with the combined results of the study providing a present day baseline for the river’s ecological condition.
